Question 2~ Use transparent gifs or pngs. For Flash, I recommend pngs. They're a little larger (file size) coming in but are actually smaller at publishing and are better quality.
Question 1~There is a way to calibrate but my brain is a little fuzzy (must be the painkillers from the dentist) and I can't remember how right now. If nobody else answers by morning I'll get back to you.

What you're experiencing is the difference between RGB and CMYK. you work in RGB in PS to to get full function, and for the web, your printer prints in CMYK. My Epson printer manual says even if you work in CMYK to convert to RGB before printing. We had this discussion with Cappy a short while ago, search the threads it is very enlightening, since then , because of the Epson manual, I have been paying very close attention to the gamut warning in the color picker and have had excellent results matching print color and screen color. Unfortunately you can't print the bright colors you see on the screen do to the physical limitations of the ink we print with. I hope this helps.
